ok to defrag while downloading?

I doubt it will do anything tbh. You'll be trying to write to a disk that is trying to move large numbers of files around, making everything five times slower. And it won't be able to defrag any files in use, so it won't do a proper job. That's if it doesn't just keep trying to restart the defrag because the disk's being used.
 
I sometimes leave my PC on overnight to download large files. Would it be ok to defrag my pc at this time as well?

Depends on the defrag app. If it's got half a brain, it'll back off a disk that's in use (especially with torrents constant read/write) and defrag only when the drive is idle. If the defragger is dumb, it'll try to defrag when the torrent is running, and cause disk thrashing.:p

I run my Diskeeper 2009 Pro on auto all the time, including for the torrent drive, and it never interferes until the torrents are done, only then does it resume defrag. Same with any other app that uses resources- DK gives those priority- always so there is no problem at all.

If you use utorrent, you can pre-allocate the space for the files, so they don't fragment..ofcourse under the assumption that sufficient contiguous free disk space is already available.
