Partitioning does not really affect speed (as long as they are not too small), but which part of the HDD is being read from, the outer part will be the fastest, the innermost, slowest, due to how far the read/write heads have to travel across the HDD to access data.
A partitioning example:
2 x 74GB WD Raptors in RAID0, 148GBs, C:\ - 20-30GBs from XP-Vista, remaining space, leave as it is, and make D:\, or split into 2 equal spaces, put My Documents onto D:\, or some people do not partition at all, really a personal preference it seems.
