Say I’ve got one 400gb HDD and I partitioned it into say one 100gb section for OS and progs and another 300gb section for storage music, video, ect.

Would a partitioned drive run slower or be any less reliable than say having a dedicated OS and progs drive and a separate Storage drive??

Partitioned drive - only one read / write head in use.

Multiple drives - multiple read / write heads in use.

Or - look at what happens when you copy a file from one hard disk to another. And then compare this with the time it takes to copy one file to another folder on the same drive.

Two drives faster than one. :)

I have multiple partitions, but mainly to keep things tidy and reduce fragmentation. I still off load tasks like swap files and temps files to separate hard drives.
