This happens in populations already. I think a certain seal population was affected and that was the example we were given in uni. It's called the Bottleneck Effect and occurs when a species is near extinction but starts to recover but now the whole species is gentically similar and so won't adapt very well.

On the other hand Genetic Drift occurs more frequently in small populations so the population can change over time.

Also, the Amish amd Afrikaners established new populations from small numbers of individuals (The Founder Effect).

Unfortunately we were not given actual numbers needed for a population to continue so I can't answer your question directly but with the Amish they can be traced back to a single couple.
 
We've never been inbred, we're the result of evolution... prior to being mamals we'd have been some kind of fish or something (I don't read up on evolution much ok?) and there would have been thousands of us forming into primates at the same time. As far as I know when you're a fish it's cool to inbreed.
