Drive difference

I have been looking at getting a new drive for my system, this will be my boot drive. I have most of my installed programs (games etc on a seperate drive) and wondered what is best to go for.

New drive just for OS and 2nd drive for Games

or

New drive for OS and a partition for storage.

Which would be the quicker and best option?

Been looking at these 2 drives and wondered what the difference is apart from size. Also which is best choice?

Samsung SpinPoint F1 250GB SATA-II 16MB Cache - OEM (HD252HJ)

Samsung SpinPoint F1 320GB SATA-II 16MB Cache - OEM (HD322HJ)
 
Ok thanks everyone for the advice, i will go for the 320g F1 version.

Now all i need to know if how to use it as 1 boot drive or partitioned? Also will it get dramatically slower the more the drive gets filled?

Samsung SpinPoint F1 640GB

There are a few posts in the forum that say there is but i wondered if this is true as they both use 320gig platters.
