Prepare! Do your research extensively and have good, solid answers to the questions they are most likely to ask (they're usually very similar each time). Do some mock interviews.
I don't really feel that breathing techniques and the like will assist too much. Once you're in the hot seat, you either know what you're doing and therefore feel confident or you don't. Taking a few deep breaths beforehand won't really change this!
I had a big interview last week for a senior managerial position, I had a 700 mile round trip and got up at 3am for my 10:30 interview, on the drive down this gave me plenty of time to think about it! Best advice I can give is, they can only say NO, be yourself(or your sis to be herself) and try to RELAX! They know people are going to be nervous and expect some wayward answers when people try to OVER IMPRESS!
BTW I was offered the job but declined it in the end
