Right. Manchester Piccadilly to Media City. City centre, so you may find shortcuts depending on your own personal feelings about, you know, one way signs and the like. Google Maps cycling directions offers some interesting ideas as well, which you may want to consider working in - little cobbled sections might liven things up, for example.
I think I'd probably go for something like this. You could substitute Whitworth Street for Portland Street on the first half of the journey, but it's a faff getting onto Whitworth Street from Piccaddilly unless you fancy doing a u-turn to get onto it or going down to Fairfield Street and getting across to Whitworth that way. Either way I'd recommend picking up Liverpool Road once you cross the A56 and heading across the bridge and down the little cycle path at the end to Middlewood Street and Liverpool Street. You could use the A57 instead of Liverpool Street, but the A57 is busy dual carriage way and Liverpool Street isn't. Both have cycle paths for at least part of the routes, though. The junction to get from the A6042 onto the A57 is pretty silly though, and while there are cycle paths to get across it, you could end up waiting a fair bit of time for the pelican crossings.

Brilliant. Someone had mentioned Princes Bridge to me and I was looking at a very similar route to that one. Since I'll be on the Brompton I might just walk it over to Whitworth and start there.
I usually get a cab too and from the ariport (but have to get the train now) so I've seen folks cycling the A57 and it looks OK, but only issue is that some of the roundabouts and junctions look a nightmare.
